چکیده

Tumors contain a sub-population of self-renewing and expanding cells known as cancer stem cells (CSCs). Putative CSCs were isolated from human melanoma cells of a different aggressiveness, Hs294T and A375 cell lines, grown under hypoxia using "sphere-forming assay", CD133 surface expression and migration ability. We found that a cell sub-population enriched for P1 sphere-initiating ability and CD133 expression also express larger amount of VEGF-R2. Etoposide does not influence phenotype of this sub-population of melanoma cells, while a combined treatment with Etoposide and Bevacizumab significantly abolished P1 sphere-forming ability, an effect associated with apoptosis of this subset of cells. Hypoxic melanoma cells sorted for VEGF-R2/CD133 positivity also undergo apoptosis when exposed to Etoposide and Bevacizumab. When Etoposide and Bevacizumab-treated hypoxic cells were injected intravenously into immunodeficient mice revealed a reduced capacity to induce lung colonies, which also appear with a longer latency period. Hence, our study indicates that a combined exposure to Etoposide and Bevacizumab targets melanoma cells endowed with stem-like properties and might be considered a novel approach to treat cancer-initiating cells.

Expression of Recombinant Coagulation Factor IX in Human Amniotic Membrane-derived Mesenchymal Stem Cells: A New Strategy to Gene Therapy of Hemophilia B

Background: Hemophilia B is an X-linked hereditary disorder of blood coagulation system which is caused by factor IX (FIX) deficiency. Factor IX is a plasma glycoprotein that participates in the coagulation process leading to the generation of fibrin. Replacement of factor IX with plasma-derived or recombinant factor IX is the conventional treatment for hemophilia B to raise the factor IX le...

Evaluation of In Vitro Differentiation of Cardiomyocyte-like cells Derived from Human Bone Marrow Mesenchymal Stem Cells

Purpose: To investigate the in vitro differentiation process of cardiomyocyte-like cells derived from human bone marrow mesenchymal stem cells under the influence of 5-azacytidine (5-aza). Materials and Methods: After purification, human bone marrow mesenchymal stem cells were exposed to 5-aza at a concentration of 5 μmol for 5 weeks to induce cardiomyocyte differentiation. To induce differenti...

Melanoma, B16-F10 Protective Immunity against Highly Metastatic Mouse Glypican-3, a Recently Identified Oncofetal Antigen, Induce Derived Dendritic Cells

We have recently established a method to generate dendritic cells from mouse embryonic stem cells. By introducing exogenous genes into embryonic stem cells and subsequently inducing differentiation to dendritic cells (ES-DC), we can now readily generate transfectant ES-DC expressing the transgenes. A previous study revealed that the transfer of genetically modified ES-DC expressing a model anti...

Functional Inhibition of Nucleostemin Gene-Acoordinator of Self-Renewal Ability-In Bone Marrow Derived Mesenchymal Stem Cells by Rnai Strategy

Purpose: The aim is to downregulate the expression level of NS as an important factor in sustaining stem cells and certain types of cancer cells self-renewal ability in bone marrow derived mesenchymal stem cells by RNAi strategy and investigate the effects of absence of NS in these cells. Materials and Methods: Double strand NS-specific and control siRNA oligos were designed and transfected in...

Differentiation of Mesenchymal Stem Cell toward the Insulin-like Cells with Lentivirus Vector Mir-375

Background & Objective: Type1 diabetes is characterized by autoimmune destruction of pancreatic β cells, leading to reduced insulin secretion. Differentiation of mesenchymal stem cells (MSCs) into β-like cells offers new ways of diabetes treatment. MSCs can be insulated from the human umbilical cord tissue and differentiate into insulin-producing cells. Material & Methods: Human um...
